Since it is in Stirling Castle, I was thinking it might be one of the Jameses, maybe James I.

James had two legitimate sons who dies as infants - I will need to get home to look up dates for them. The elder, James, lived long enough to be still alive when his brother, Robert, was born.
The child could not be Mary Queen of Scots as she was barely a week old at James V's death in December 1542.

As he said, "it (the Stewart dynasty) came wi' a lass (Marjorie Bruce) and it will pass wi' a lass (Mary)". Technically Darnley's surname ought to have prevailed on her deposition but it didn't any more than it will when the present "lass" passes.
The portrait looks like a boy so it could be his elder son - I think Robert caught a fever, passed it to James and both died - an illegitimate child of James V or an older portrait.

To answer my own question - in fact to correct myself - Darnley's surname was STUART and his wife's STEWART, explaining why the next dynasty in England had a variant on the name!
