It seems to me, working out the maternal line, that this mitochondrial DNA they are using for these tests goes back to Katherine Swynford: Cecily Neville, Richard's mother, was the daughter of Joan, who in turn was the daughter of Katherine and John of Gaunt. a Beaufort!
and does anyone have any suspicions or info regarding the identity of the second anonymous DNA donor?

This is an exceedingly secure link. mtDNA is inherited only through the female line so infidelity by one or more of these wives would have no effect. Katherine de Roet would indeed be a few places further up.
